Thesis title: “Identification of the Herbal Homeopathic Formulations with Anti-Cervical Cancer and Anti-HPV Activity”.
Salient features of Ph.D. research work
The present study carries out the comprehensive evaluation of anti-CaCx and anti-HPV activities of certain plant-derived preparations (HP) which supports their repurposing in the management of CaCx as economical alternatives. HP mother tincture (MT) and 30C potencies of plants Berberis aquifolium (BA), Berberis vulgaris (BV), Mentha piperita (MP), Curcuma longa (CL), Cinchona officinalis (CO), Thuja occidentalis (TO) and Hydrastis canadensis (HC) were screened for their anti-proliferative activity on human papillomavirus (HPV)-negative (C33a) and HPV-positive CaCx cells (SiHa and HeLa) by MTT assay. These plant-derived HPs showed a multi-pronged therapeutic potential against HPV infection and cervical cancer and described the underlying molecular mechanism of their action and the role of transcription factors like AP-1 and STAT-3 in the disease pathogenesis. Thus, these phytomedicines can potentially meet the clinical need in the form of first-line or adjunct therapy for cervical cancer treatment. Factors like AP-1 and STAT-3 in the disease pathogenesis. I, Dr. Tejveer Singh worked on “Development of Anti-HPV and Anti-Cervical Cancer Therapeutic”. Briefly, I studied the active action of plant-derived different formulations on HPV-associated in cervical cancer cell line (CaCx) models (in vitro and in silico). We observed anti-proliferative activity, cell cycle arrest in the G0/G1 phase with E6 and E7 of HPV 16 and 18 oncoproteins was maximally downregulated. Further, checked the phytochemicals mechanism of action at the molecular level and observed modulated expression of different transcription factor proteins such as STAT3 and AP-1 of CaCx in human cell lines. This part of the work published in Frontiers in Pharmacology and Homeopathy Elsevier journal Journal. Ph.D. thesis review work has been published in Frontier in Bioscience journal.
I am well equipped with therapeutic research skills which include their biophysical cytotoxic assay: MTT assay, molecular characterization methods (DNA, RNA and protein extraction, PCR, western blotting, fluorescence microscopy, and confocal microscopy), analyzing of cell cycle and apoptosis (FACS), Protein purification, Animal handing, Patient derived xenograft model, Human and animal Tissue culture (2D and 3D model System) and antioxidant assays (TPC, DPPH, and ABTS assay). I am also learned different computation biology tools (AutoDock, PyMol, UniProt, and Discovery studio) and cytogenetics from different collaborative cancer research institutes.
Apart from this, I have worked on different projects and cancer model like as head and neck cancer study was published in Cancer Medicine and Frontiers in Cellular and Infection Microbiology, prostate cancer research work published in Journal of Cellular Biochemistry, stem cancer cell project work published in Tumor Biology and Scientific Reports, exosome as a diagnostic tool in cancer model published in Cancer Cell International.
